In this section, Matthew Arango, PharmD, outlines how treatment for HER2-overexpressed gastroesophageal adenocarcinoma (GEA) has traditionally been approached and what distinguishes the major HER2-targeted agents used or emerging in this space. He discusses the pharmacologic differences between trastuzumab (Herceptin; Genentech), trastuzumab deruxtecan (Enhertu; Daiichi Sankyo, AstraZeneca), and zanidatamab (Ziihera; Jazz Pharmaceuticals). Arango also walks through the key findings from the HERIZON-GEA-01 study evaluating zanidatamab, noting that this triplet regimen remains investigational as of late February 2026.
